Item Antibacterial activity of extract and fractions from branches of Protium spruceanum and cytotoxicity on fibroblasts.(2017) Amparo, Tatiane Roquete; Rodrigues, Ivanildes Vasconcelos; Seibert, Janaína Brandão; Souza, Rafaella Hilda Zaniti; Oliveira, Amanda Ribeiro de; Cabral, Vivette Appolinário Rodrigues; Vieira, Paula Melo de Abreu; Brandão, Geraldo Célio; Okuma, Adriana Akemi; Vieira Filho, Sidney Augusto; Teixeira, Luiz Fernando de Medeiros; Souza, Gustavo Henrique Bianco deThe crude ethanol extract (CEE) and fractions from branches of Protium spruceanum were subjected to antibacterial and cytotoxicity assays. Compounds of the most active fraction were identified by GC-MS and LC-MS. CEE was active against 19 bacteria and the ethyl acetate fraction (EAF) showed the lowest minimum bactericidal concentration (MBC 0.3–80.0 mg/mL). Through time-kill assay was observed that EAF induced rapid bactericidal effect against Staphylococcus saprophyticus. The cytotoxicity tests against L929 fibroblasts showed great potential of EAF on the treatment of infections caused by five bacteria (MBC < IC50). The results provide in vitro scientific support to the possible application of branches of P. spruceanum as antimicrobial agent that may contribute for treatment of infections.Item Antimicrobial activity of synthetic bornyl benzoates against Trypanosoma cruzi.(2012) Corrêa, Paulo Roberto Ceridóreo; Miranda, Roqueline Rodrigues Silva de; Duarte, Lucienir Pains; Silva, Grácia Divina de Fátima; Vieira Filho, Sidney Augusto; Okuma, Adriana Akemi; Carazza, Fernando; Díaz, José Andrés Morgado; Pinge Filho, Phileno; Yamauchi, Lucy Megumi; Nakamura, Celso Vataru; Ogatta, Sueli Fumie YamadaWe report here for the first time the in vitro effects of (1S,2R,4S)-1,7,7-trimethyl-bicyclo[2.2.1]heptan-2-yl- 39,49,59-trimethoxy benzoate (1) and (1S,2R,4S)-1,7,7-trimethyl-bicyclo[2.2.1]heptan-2-yl benzoate (2) on the growth and ultrastructure of Trypanosoma cruzi. These two synthetic compounds exerted an antiproliferative effect on the epimastigote forms of the parasite. The ICs50/72h of two synthetic L-bornyl benzoates, 1 and 2, was 10.1 and 12.8 mg/ml, respectively. Both compounds were more selective against epimastigotes than HEp-2 cells. Ultrastructural analysis revealed intense cytoplasmic vacuolization and the appearance of cytoplasmic materials surrounded by membranes. The treatment of peritoneal macrophages with compounds 1 and 2 caused a significant decrease in the number of T. cruzi-infected cells. L-Bornyl benzoate derivatives may serve as a potential source for the development of more effective and safer chemotherapeutic agents against T. cruzi infections.Item Evaluation of the, in vitro, photoprotetive capacity of Moringa oleifera oil for its use in sunscreen formulation.(2021) Mota, Gabriela Cristina Ferreira; Campos, Kamila de Fátima Anunciação; Sousa, Lucas Resende Dutra; Amparo, Tatiane Roquete; Vieira, Paula Melo de Abreu; Okuma, Adriana Akemi; Melo, Tânia Márcia Sacramento; Santos, Viviane Martins Rebello dosMoringa oleifera Lam is an Indian plant with applications in the agricultural and medical fields. The assets development capable of increasing the efficiency of sunscreens, mainly those of plant origin, due to their natural benefits, represents an increasing demand for cosmetology. The present study aims to identify by CG-MS the constituents of the most active oil and to evaluate the photoprotective capacity of Moringa oil, and its action in sunscreen formulations. Extracts of the oils from the Moringa seeds were evaluated for the Sun Protection Factor (SPF) observing the highest result for the dichloromethane extract. This extract showed low cytotoxic potential for human fibroblasts and it was incorporated into a sunscreen. The extract increased the SPF of the sunscreen and its effect may be related to fatty acids identified by GC-MS. The results showed the benefit of Moringa oil as a vegetable active in the sunscreen formulations by increasing the SPF of sunscreens in a natural and sustainable way.Item Influence of environmental factors and cultural methods on the content of N,N-Dimethyltryptamine in Psychotria viridis (Rubiaceae).(2018) Cavalcanti, André Dias; Cardoso, Gabriele de Azevedo; Oliveira, Fernando Luiz Pereira de; Bearzoti, Eduardo; Okuma, Adriana Akemi; Duarte, Lucienir Pains; Vieira Filho, Sidney AugustoPsychotria viridis is one of the species that produces N,N-dimethyltryptamine. Its decoction together with other species, such as Banisteriopsis caapi, produces ayahuasca, a beverage used for ritualistic and medicinal purposes. The goal of this study was to understand how environmental factors and cultivation methods influenced the content of N,N-dimethyltryptamine in P. viridis. Over all four seasons, leaf samples were collected from 25 different locations in 14 Brazilian states, and Federal District. Environmental parameters, micro and macronutrients, plant characteristics, information on farming methods were correlated with N,N-dimethyltryptamine content, determined by gas chromatography coupled to mass spectrometry (GC-MS). Greatest effects on the N,N-dimethyltryptamine amount were associated with seasonality, altitude, latitude and biome type. A positive correlation between N and Mg content and N,N-dimethyltryptamine levels was statistically established. By regression analysis, the adequate foliar nutrient levels that would result in the concentration of N,N-dimethyltryptamine in cultivated plants similar to that of Amazonian P. viridis were equated.
