    Angiotensin‐converting enzyme gene (ACE) polymorphisms are associated with dysregulation of biochemical parameters in hypertensive patients.

    Introduction The genetic component, including genes and their variants, plays a signifcant role in the pathophysiology of arterial hypertension (AH). Thus, clinical, epidemiological and genetic studies have been carried out to improve the under- standing of disease mechanisms, improve diagnostic quality and contribute to prevention. Objective To determine the association of risk factors, biochemical parameters and diferent ACE gene polymorphisms with AH. Method The case-control study was carried out in the population of Ouro Preto, Brazil. The subjects answered a question- naire containing clinical and sociodemographic data. The ACE gene polymorphisms rs4291, rs4363 and rs4335 were evalu- ated by real time-polymerase chain reaction (real-time PCR) in 310 people (155 hypertensive and 155 normotensive patients), in addition to biochemical parameters. A multivariate logistic regression model was used to identify factors associated with AH. Analysis of continuous variables was performed using the Kruskal-Wallis test to assess signifcance between groups and Dunn’s post-test for multiple comparisons. Results The results showed that AH was associated with age, education, smoking, obesity and high levels of triglycerides, sodium, glucose and uric acid. Regarding the biochemical parameters, in hypertensive patients, the rs4363 and rs4335 poly- morphisms were associated with high levels of triglycerides, urea and glucose; the rs4291 polymorphism was associated with elevated urea and glucose levels. No association was detected between SNPs and HA. Conclusion AH was associated with socioeconomic status, lifestyle habits and biochemical parameters. ACE polymorphisms may have infuenced the levels of triglycerides, urea and glucose in hypertensive patients.

    Persistence or clearance of human papillomavirus infections in women in Ouro Preto, Brazil.

    Persistent high-risk (HR) human papillomavirus (HPV) infection is necessary for development of precursor lesions and cervical cancer.We investigate persistence and clearance of HPV infections and cofactors in unvaccinated women. Cervical samples of 569 women (18–75 years), received for routine evaluation in the Health Department of Ouro Preto, Brazil, were collected and subjected to PCR (MY09/11 orGP5+/6+ primers), followed by RFLP or sequencing. All women were interviewed to collect sociodemographic and behavioral information. Viral infection persistence or clearance was reevaluated after 24 months and was observed in 59.6% and 40.4% of women, respectively. HPVs 16, 33, 59, 66, 69, and 83 (HR) were the most persistent types whereas HPVs 31, 45, and 58 were less persistent. Clearance or persistence did not differ between groups infected by HPVs 18, 53, and 67. In low-risk (LR) types, HPV 6 infected samples were associated with clearance, while HPV 11, 61, 72, or 81 infected samples were persistent in the followup. No statistically significant association was detected between persistent HPV infections and sociodemographic and behavioral characteristics analyzed. To study persistence or clearance in HPV infection allows the identification of risk groups, cofactors, and strategies for prevention of cervical cancer.

    Effect of substrate and pH on the activity of proteases from Fusarium oxysporum var. lini.

    The results obtained in this work suggest that both the pH (through selective inhibition) and the carbon source (through repression and acidification or alkalinization of the medium) may play an important role in the distribution of extracellular proteases in Fusarium oxysporum var. lini.

    Morphometric evaluation and nonclassical criteria for the diagnosis of HPV infection and cytological atypia in cervical samples.

    Herein, we evaluated cervical samples from normal tissue or HPV-infected tissue, to determine if the relative nuclear/cytoplasmic ratio (NA/CA) and the presence of nonclassical cytological criteria are a novel cytological criterion for the diagnosis of HPV. Significantly, larger NA/CA ratios were found for the HPV−ATYPIA+ and HPV+ATYPIA+ groups compared with HPV−ATYPIA− group, regardless of collection method. For the samples collected with a spatula, only three samples from the HPV−ATIPIA− group showed four or more nonclassical parameters (i.e., were positive), while a larger number of the samples in the HPV−ATYPIA+, HPV+ATYPIA−, and HPV+ATYPIA+ groups were positive (13, 4, and 13 samples, respectively). Among those collected with a brush, no sample showed four or more nonclassical criteria in the HPV−ATYPIA− group, while a number of samples were positive in the HPV−ATYPIA+, HPV+ATYPIA−, and HPV+ATYPIA+ groups (4, 3, and 4 samples, respectively). HPV infection was associated with significant morphometrical changes; no increase in the NA/CA ratio was found in the HPV+ATYPIA- samples, compared with the HPV−ATIPIA− samples collected with either a spatula or a brush. In conclusion, by including nonclassical cytological criteria into the patient diagnosis, we were able to reduce the number of false negative and false positive HPV diagnoses made using conventional cytology alone.     Quality evaluation of simvastatin compounded capsules.

    No Brasil, a sinvastatina está comercialmente disponível na forma de comprimidos e cápsulas manipuladas. Poucos relatos estão disponíveis sobre a qualidade e, consequentemente, a eficácia dessas cápsulas. A qualidade de 30 lotes de sinvastatina 20 mg cápsulas do mercado foi avaliada através da determinação de peso, uniformidade de conteúdo, desintegração (Farmacopéia Brasileira), doseamento e teste de dissolução (monografia comprimidos USP32). Método por CLAE foi desenvolvido para o doseamento, uniformidade de conteúdo e teste de dissolução; além disso, especificações foram estabelecidas. Dos 30 lotes avaliados, 29 apresentaram desintegração da cápsula até 45 min e a variação do peso individual foi ± 10% ou ± 7,5% em relação ao peso médio, se ≤ ou > 300 mg, respectivamente. Apenas 27 lotes preencheram os critérios do teste de dissolução com valores ≥ 80% da quantidade rotulada, em 45 min, 21 lotes apresentaram conteúdo de sinvastatina entre 90,0-110,0% do valor rotulado e 19 lotes apresentaram pelo menos 9 em 10 cápsulas, com valores de uniformidade de conteúdo entre 85,0 -115,0% da quantidade rotulada com RSD ≤ 6,0%. Apenas 14 de todos os lotes (30) atenderam completamente os padrões de qualidade farmacopéicos. O estabelecimento das condições para os testes e de especificações para os parâmetros das cápsulas de sinvastatina mostrou que houve diferenças relevantes na qualidade farmacopeica entre os lotes das cápsulas manipuladas por distintas farmácias. A eficácia do tratamento hipercolesterolêmico poderia estar comprometida para 53,33% dos produtos testados.

    Self-similarities of periodic structures for a discrete model of a two-gene system.

    We report self-similar properties of periodic structures remarkably organized in the two-parameter space for a two-gene system, described by two-dimensional symmetric map. The map consists of difference equations derived from the chemical reactions for gene expression and regulation. We characterize the system by using Lyapunov exponents and isoperiodic diagrams identifying periodic windows, denominated Arnold tongues and shrimp-shaped structures. Period-adding sequences are observed for both periodic windows. We also identify Fibonacci-type series and Golden ratio for Arnold tongues, and period multiple-of-three windows for shrimps.

    Can established cultured papilloma cells harbor bovine papillomavirus?

    Papillomaviruses have been reported to be very dif¬ficult to grow in cell culture. Also, there are no descriptions of cell cultures from lesions of bovine cutaneous papillomatosis, with identification of different bovine papilloma virus (BPV) DNA se¬quences. In the present report, we describe primary cell cultures from samples of cutaneous lesions (warts). We investigated the si¬multaneous presence of different BPV DNA sequences, comparing the original lesion to different passages of the cell cultures and to peripheral blood. BPV 1, 2 and 4 DNA sequences were found in lesion samples, and respective cell cultures and peripheral blood, supporting our previous hypothesis of the possible activity of these sequences in different samples and now also showing how they can be maintained in different passages of cell cultures.

    Simvastatin assay and dissolution studies by feasible rp-hplc in tablets.

    Commonly used HPLC acetonitrile solvent has been through a worldwide shortage with a cost increase in 2008 and 2009. In order to get around this situation, a method by RP-HPLC employing methanol and aqueous acid mobile phase was developed and validated to evaluate simvastatin. The quality control assay and dissolution studies of this lipid-lowering drug were performed in diluents methanol and 0.01 M phosphate buffer with 0.5% SDS, pH 7, respectively. Dissolution test aliquots did not go through sample treatment, as described in USP SIM tablets monograph by ultraviolet spectrophotometry. The proposed method is fast, simple, feasible and robust.

    Influência da temperatura em estudos de dissolução de formas farmacêuticas contendo sinvastatina.

    O antihiperlipêmico sinvastatina está disponível no mercado nacional na forma de comprimidos e cápsulas manipuladas. Método por cromatografia a líquido de alta eficiência com detecção no ultravioleta (l 238 nm) foi utilizado para quantificar sinvastatina em cápsulas e comprimidos em tampão fosfato de sódio monobásico 0,01 M contendo laurilsulfato de sódio 0,5%, pH 7, utilizado como diluente ou meio de dissolução. A estabilidade de curto prazo do fármaco das amostras foi avaliada no diluente, mantido a diferentes temperaturas. A estabilidade de curto prazo (até 24 h) de soluções de sinvastatina, não submetidas ao teste de dissolução, foi satisfatória para o fármaco isolado e para cápsulas, o que não se verificou para comprimidos. A estabilidade da sinvastatina em soluções mantidas a 22-24 ºC por 8 h, após o teste de dissolução foi satisfatória para o fármaco isolado e para cápsulas contendo butilhidroxitolueno como excipiente. Os resultados de amostras de sinvastatina cápsulas contendo vitamina B12 e de comprimidos indicam a necessidade de refrigeração das soluções em estudos de dissolução, caso a análise cromatográfica tenha duração superior a 4 h e 2 h, respectivamente, como descrito.

    A qualidade, o custo e a dispensação de sinvastatina cápsulas em farmácias magistrais.

    Com base na RDC/ANVISA 67/2007, avaliou-se o serviço prestado na dispensação de sinvastatina cápsulas em farmácias magistrais. Vinte e quatro (16%) das 150 farmácias magistrais foram aleatoriamente selecionadas nas regiões de Belo Horizonte-MG, observando a distribuição geográfica. A qualidade do produto, os aspectos da dispensação e o custo foram considerados em estudos de correlação. Os resultados foram submetidos a análises estatísticas descritivas e inferenciais. Em apenas 25% destas farmácias, o fax da prescrição foi solicitado. Todos os pedidos foram entregues no prazo, porém sem comprovação da dispensação. Em uma das entregas, o medicamento era o incorreto. Seis (25%) dos produtos apresentaram rotulagem incompleta (ausência de posologia 16,3% ou de identificação do médico/paciente 8,7%). Apenas 11 (45,8%) das amostras apresentaram qualidade satisfatória. O custo do medicamento manipulado variou entre R$20,40 e R$44,00 com valor médio R$31,18. Não se observou associação entre a qualidade do medicamento e os aspectos do custo, prazo de entrega e localização geográfica. Verifica-se que a maioria das farmácias magistrais ainda precisa adequar-se à legislação para garantir à população uma assistência farmacêutica de qualidade.
