Item Avaliação imunopatológica do tecido muscular cardíaco canino, submetido ao efeito da Azatioprina e infecção por diferentes formas da cepa Be-78 do Trypanosoma cruzi, durante a fase aguda.(2014) Souza, Sheler Martins de; Carneiro, Cláudia MartinsAs duas formas de infecção pelo Trypanosoma cruzi, tripomastigota metacíclica (TM) e sanguínea (TS) são distintas em relação ao processo de interação com as células do hospedeiro, devido às moléculas presentes em sua superfície. Esta diferença pode desencadear diferente resposta imunológica. Resposta esta, que também pode ser alterada com o tratamento pelo fármaco Azatioprina. Com base nestes aspectos, o objetivo deste trabalho foi avaliar a resposta imune no coração de cães tratados com a Azatioprina ou não, durante a fase aguda da infecção por formas TM ou TS da cepa Berenice-78 do T. cruzi. Ao avaliar o parasitismo e a resposta imune no tecido muscular cardíaco de cães infectados por formas TM, verifica-se que estes apresentaram um aspecto histopatológico mais preservado, com baixo parasitismo, um balanço favorável, evidenciado pela razão, da expressão da IL-12p40 e uma maior expressão dos receptores TLR2 e TLR4. Em contrapartida, os cães infectados por formas TS apresentaram destruição tecidual, alto parasitismo, intenso processo inflamatório e elevada expressão das citocinas, quimiocinas, CCR5 e TLR9. Nos animais submetidos à infecção por diferentes formas do T. cruzi e tratados com a Azatioprina, ocorreu um aumento do parasitismo em ambos os grupos (TMA e TSA) e do processo inflamatório no grupo TMA, sendo estes fatores determinantes na gênese das lesões da fibra muscular cardíaca. Além disso, pode-se constatar aumento das citocinas TNF-α, IFN-γ, IL-4, IL-10 e TGF-β1, da quimiocina CCL3/MIP-1α e TLR9 no grupo TMA. Em suma, a ação da Azatioprina parece conduzir à resposta imune distinta. Esta evidência foi observada em ambas as formas de infecção, culminando em uma piora dos aspectos histopatológicos, principalmente naqueles do grupo TMA. Este fato pode estar ligado a mecanismos co-estimulatórios, via CD28. Sabe-se que a Azatioprina pode levar ao bloqueio desta via que é de grande importância para desencadear uma resposta imune Th1. Neste sentido, o bloqueio ou mesmo o desequilíbrio desta via e, consequentemente, da resposta imune de cunho protetora, favorece a replicação parasitária. Sugere-se a possível importância da via CD28 nos mecanismos de defesa contra o T. cruzi e chama-se a atenção para a função dos receptores TLR2 e TLR4 na infecção chagásica, pois estes podem desempenhar função protetora.Item Caracterização imuno-histoquímica do infiltrado inflamatório cardíaco durante a fase aguda da infecção experimental de cães por formas tripomastigotas metacíclicas ou sanguíneas da cepa Berenice-78 do Trypanosoma cruzi.(Programa de Pós-Graduação em Ciências Biológicas. Núcleo de Pesquisas em Ciências Biológicas, Pró-Reitoria de Pesquisa e Pós Graduação, Universidade Federal de Ouro Preto., 2010) Souza, Sheler Martins de; Carneiro, Cláudia MartinsNosso grupo vem estudando a infecção experimental de cães pelo Trypanosoma cruzi ao longo dos últimos 30 anos. Semelhante ao observado em humanos, no modelo canino verifica-se um infiltrado inflamatório predominantemente constituído por células mononucleares durante a fase aguda da doença. A identificação destas células é uma estratégia importante para a compreensão dos mecanismos imunopatológicos desencadeados pelo parasito ao longo da infecção. Neste contexto, fez-se necessário a padronização de reações imuno-histoquímicas para caracterização do fenótipo das principais células presentes no infiltrado inflamatório (linfócitos T CD4 + e T CD8 + , macrófagos CD14 + e neutrófilos). As células envolvidas na infecção experimental aguda de cães por formas tripomastigotas metacíclicas ou sanguíneas da cepa Berenice-78 do T. cruzi foram quantificadas por análise morfométrica, no parênquima e no foco inflamatório, tanto no átrio direito quanto no septo interventricular. Observou-se que em ambos os fragmentos cardíacos, o processo inflamatório foi maior no grupo infectado por formas tripomastigotas sanguíneas. A quantificação do fenótipo celular mostrou que os linfócitos T CD4 + são predominantes no foco inflamatório nos animais infectados por ambas as formas, tanto no átrio direito quanto no septo interventricular. No átrio direito, os linfócitos T CD8 + foram observados em maior quantidade nos grupos infectados em relação ao grupo controle. Já os macrófagos CD14 + prevalecem apenas no grupo infectado por formas tripomastigotas metacíclicas em relação ao grupo controle. No septo intraventricular, os linfócitos T CD8 + e os macrófagos CD14 + encontram-se em elevado número no grupo infectado por formas tripomastigotas metacíclidas em relação ao grupo controle e ao grupo infectado por formas tripomastigotas sanguíneas. Os neutrófilos estão presentes em menor número em relação às demais células analisadas, mas são encontrados em maior número no átrio direito de ambos os grupos infectados. O quadro histopatológico mais preservado observado no grupo infectado por formas tripomastigota metacíclico pode estar diretamente relacionado à interação entre os linfócitos T CD8 + e os macrófagos CD 14 + , pois estas são potentes produtoras de interleucina-12, uma citocina que estimula a produção de interferon-gama pelos linfócitos T CD8 + . Esta citocina é fundamental para o processo de resistência à infecção pelo T. cruzi durante a fase aguda por estimular a síntese de óxido nítrico pelos macrófagos. A quantificação celular realizada de modo aleatório evidenciou melhor as diferenças entre os grupos. Contudo, a quantificação focal do processo inflamatório distinguiu melhor as porcentagens das populações e subpopulações celulares. Os resultados obtidos mostraram que as formas infectantes, tripomastigotas metacíclicas ou sanguíneas, apresentam perfil celular distinto no átrio direito e no septo intrerventricular, sendo a resposta elaborada durante a infecção com as formas tripomastigotas metacíclicas mais eficiente e menos lesiva ao hospedeiro.Item Development of chronic cardiomyopathy in canine Chagas disease correlates with high IFN-g, TNF-a, and low IL-10 production during the acute infection phase.(2009) Guedes, Paulo Marcos da Matta; Veloso, Vanja Maria; Afonso, Luís Carlos Crocco; Caliari, Marcelo Vidigal; Carneiro, Cláudia Martins; Diniz, Lívia de Figueiredo; Silva, Eduardo de Almeida Marques da; Caldas, Ivo Santana; Matta, Maria Adelaide do Valle; Souza, Sheler Martins de; Lana, Marta de; Chiari, Egler; Galvão, Lúcia Maria da Cunha; Bahia, Maria TerezinhaWhen infected with Trypanosoma cruzi, Beagle dogs develop symptoms similar to those of Chagas disease in human beings, and could be an important experimental model for a better understanding of the immunopathogenic mechanisms involved in chronic chagasic infection. This study evaluates IL-10, IFN-g and TNF-a production in the sera, culture supernatant, heart and cervical lymph nodes and their correlation with cardiomegaly, cardiac inflammation and fibrosis in Beagle dogs infected with T. cruzi. Pathological analysis showed severe splenomegaly, lymphadenopathy and myocarditis in all infected dogs during the acute phase of the disease, with cardiomegaly, inflammation and fibrosis observed in 83% of the animals infected by T. cruzi during the chronic phase. The data indicate that infected animals producing IL-10 in the heart during the chronic phase and showing high IL-10 production in the culture supernatant and serum during the acute phase had lower cardiac alterations (myocarditis, fibrosis and cardiomegaly) than those with high IFN-g and TNF-a levels. These animals produced low IL-10 levels in the culture supernatant and serum during the acute phase and did not produce IL-10 in the heart during the chronic phase of the disease. Our findings showed that Beagle dogs are a good model for studying the immunopathogenic mechanism of Chagas disease, since they reproduce the clinical and immunological findings described in chagasic patients. The data suggest that the development of the chronic cardiac form of the disease is related to a strong Th1 response during the acute phase of the disease, while the development of the indeterminate form results from a blend of Th1 and Th2 responses soon after infection, suggesting that the acute phase immune response is important for the genesis of chronic cardiac lesions.Item Dogs infected with the blood trypomastigote form of Trypanosomacruzi display an increase expression of cytokines and chemokines plusan intense cardiac parasitism during acute infection.(2014) Souza, Sheler Martins de; Vieira, Paula Melo de Abreu; Roatt, Bruno Mendes; Reis, Levi Eduardo Soares; Fonseca, Kátia da Silva; Nogueira, Nívia Carolina; Reis, Alexandre Barbosa; Tafuri, Washington Luiz; Carneiro, Cláudia MartinsThe recent increase in immigration of people from areas endemic for Chagas disease (Trypanosoma cruzi)to the United States and Europe has raised concerns about the transmission via blood transfusion andorgan transplants in these countries. Infection by these pathways occurs through blood trypomastigotes(BT), and these forms of T. cruzi are completely distinct of metacyclic trypomastigotes (MT), releasedby triatomine vector, in relation to parasite–host interaction. Thus, research comparing infection withthese different infective forms is important for explaining the potential impacts on the disease course.Here, we investigated tissue parasitism and relative mRNA expression of cytokines, chemokines, andchemokine receptors in the heart during acute infection by MT or BT forms in dogs. BT-infected dogspresented a higher cardiac parasitism, increased relative mRNA expression of pro-inflammatory andimmunomodulatory cytokines and of the chemokines CCL3/MIP-1 _, CCL5/RANTES, and the chemokinereceptor CCR5 during the acute phase of infection, as compared to MT-infected dogs. These results suggestthat infection with BT forms may lead to an increased immune response, as revealed by the cytokinesratio, but this kind of immune response was not able to control the cardiac parasitism. Infection with theMT form presented an increase in the relative mRNA expression of IL-12p40 as compared to that of IL-10or TGF- _1. Correlation analysis showed increased relative mRNA expression of IFN- _ as well as IL-10,which may be an immunomodulatory response, as well as an increase in the correlation of CCL5/RANTESand its CCR5 receptor. Beagle dogs develop the symptoms of Chagas disease in humans, and could be an important experimental model for better understanding the immunopathogenic mechanisms involved in the chagasic infection. In the present study we investigated the relation among biological factors inherent to the parasite (trypomastigote polymorphism and in vitro infectivity) and immunoglobulin production, inflammation, and fibrosis in the heart of Beagle dogs infected with either T. cruzi Y or Berenice-78 strains. In vitro infectivity of Vero cells as well as the extension of cardiac lesions in infected Beagle was higher for Y strain when compared to Berenice-78 strain. These data suggested that in vitro infectivity assays may correlate with pathogenicity in vivo. In fact, animals infected with Y strain, which shows prevalence of slender forms and high infectivity in vitro, presented cardiomegaly, inflammation, and fibrosis in heart area. Concerning the immunoglobulin production, no statistically significant difference was observed for IgA, IgM or IgG levels among T. cruzi infected animals. However, IgA together IgM levels have shown to be a good marker for the acute phase of Chagas disease.
