Item Epigenética no tecido adiposo retroperitoneal de ratos alimentados com dieta rica em carboidratos simples após treinamento físico de natação.(2018) Santos, Talita Adriana Pereira dos; Cota, Renata Guerra de Sá; Barboza, Natália Rocha; Cota, Renata Guerra de Sá; Babá, Élio Hideo; Isoldi, Mauro CésarA adiposidade corporal é um importante fator de risco para o desenvolvimento de doenças metabólicas, sugerindo um papel importante do tecido adiposo para a evolução dessas condições. Fatores ambientais como componentes dietéticos e a prática de atividades físicas podem contribuir para a modulação epigenética dos tecidos, porém não é claro o momento em que essa reprogramação acontece bem como se tais eventos antecedem o surgimento de fenótipos dessas doenças. Neste trabalho objetivamos avaliar o efeito de uma dieta rica em carboidratos simples e do treinamento físico de natação sobre a modulação epigenética no tecido adiposo retroperitoneal de animais jovens. Ratos Wistar machos recém-desmamados foram divididos em dois grupos: alimentados com uma dieta controle ou uma dieta rica em carboidratos simples e subdivididos em um grupo sedentário e um grupo treinado que foi submetido a 60 minutos de treinamento físico regular de natação com carga de trabalho por um período de quatro (20 sessões) ou oito semanas (40 sessões). Após esse período, os animais foram eutanasiados, os soros e os tecidos adiposos foram removidos para avaliar os parâmetros bioquímicos, o perfil de metilação do DNA, assim como a expressão de mRNA das enzimas metilases e desmetilases do DNA e das sirtuínas 1-7 por reação em cadeia da polimerase da transcrição reversa em tempo real. A dieta rica em carboidratos simples aumentou os coxins gordurosos retroperitoneal e epididimal, os níveis séricos de insulina e o índice HOMA-IR relacionado à hipertrofia dos adipócitos no período de 8 semanas. O treinamento físico de natação impediu o ganho excessivo de massa corporal nos animais alimentados com a dieta rica em carboidratos simples por 8 semanas, além de impedir a hipertrofia do tecido adiposo retroperitoneal induzido pela dieta. A análise comparativa revelou diminuição dos níveis de mRNA das sirtuínas 1, 2, 6 e 7 induzidos pela dieta rica em carboidratos simples e que não foi revertido pelo treinamento físico no período de oito semanas. A natação com carga de trabalho diminuiu a expressão das sirtuínas 3 e 5 no mesmo período, enquanto que a dieta rica em carboidratos simples aumentou os níveis da sirtuína 5. Em relação à metilação do DNA, não observamos alterações na metilação global, assim como na expressão gênica das enzimas envolvidas nesse processo. Assim, demonstramos que a expressão gênica de sirtuínas é modulada em função da idade e dieta e pode ser alterada através do treinamento físico, e ainda, que estas condições não induziram uma modificação na metilação do DNA nesse período de tempo estudado.Item High-sugar diet intake, physical activity, and gut microbiota crosstalk : implications for obesity in rats.(2020) Neves, Viviano Gomes de Oliveira; Oliveira, Daiane Teixeira de; Oliveira, Deborah Campos; Perucci, Luiza Oliveira; Santos, Talita Adriana Pereira dos; Fernandes, Isabela da Costa; Sousa, Graziele Galdino de; Barboza, Natália Rocha; Cota, Renata Guerra de SáThis study aims to evaluate the effect of long-term high-sugar diet (HSD) intake and regular physical activity on gut microbiota as well as its health impact. Weaned male Wistar rats were fed with standard chow diet (SSD) or HSD ad libitum and subjected or not to regular swimming training with a workload (2% of body weight) for 15 weeks. Feces samples were used on microbiome analysis using 16S rRNA amplicon sequencing. HSD increased body mass, adipose cushions, and the serum levels of triglycerides and VLDL, also changed the bacteria taxons associated with metabolic disorders (increase taxons belonging to Proteobacteria phylum and decrease Pediococcus genus); the swim training reverted these changes. SSD intake increased the abundance of bacteria associated with metabolization of dietary fiber. Training in association with SSD consumption beneficially modulated the microbiota, increasing the Bacteroidetes, Bacteroidaceae, Porphyromonadaceae, Parabacteroides, and Lactobacillaceae, and decreasing the Firmicute/Bacteroidetes ratio; training was not able to maintain this profile in animals SHD-fed. Physical training modulates the gut microbiota reversing the obesogenic response caused by SHD. However, training itself is not efficient for up-regulating the probiotic bacteria in comparison to its association with a balanced diet.Item High-sugar diet leads to obesity and metabolic diseases in ad libitum-fed rats irrespective of caloric intake.(2020) Oliveira, Daiane Teixeira de; Fernandes, Isabela da Costa; Sousa, Graziele Galdino de; Santos, Talita Adriana Pereira dos; Paiva, Nívia Carolina Nogueira de; Carneiro, Cláudia Martins; Evangelista, Elísio Alberto; Barboza, Natália Rocha; Cota, Renata Guerra de SáObjective: Provide a comprehensive view of the events surrounding the sugar consumption, under conditions of energy equivalence; through the analysis of behavioral aspects of intake, and of biochemical, metabolic and physiological parameters, as well as the effect of this nutrient on the plasticity of adipose tissue. Materials and methods: Newly weaned male Wistar rats were classified in two groups and subjected to the following normocaloric diets: standard chow diet or to high-sugar diet (HSD) ad libitum for 18 weeks. Results: The animals submitted to the HSD were associated with a lower caloric intake during the 18 weeks of experimentation. However, the HSD induced a significant increase in body weight, white adipose tissue weight, adiposity index, Lee index, and the levels of triglycerides and very low-density lipoprotein in the serum. In addition, it induced glucose intolerance, insulin resistance and compensatory increase of insulin secretion by pancreatic β-cells. Also increased heart rate and induced hyperplasia, and hypertrophy of retroperitoneal visceral adipose tissue. In the liver, the HSD was associated with increased hepatic lipid content (i.e., triglycerides and cholesterol) and hepatomegaly. Conclusion: The post-weaning consumption of HSD induces an adaptive response in metabolism; however, such an event is not enough to reverse the homeostatic imbalance triggered by the chronic consumption of this macronutrient, leading to the development of metabolic syndrome, irrespective of caloric intake. These findings corroborate recent evidence indicating that sugar is a direct contributor to metabolic diseases independent of a positive energy balance.Item Pre-eclampsia is associated with reduced resolvin D1 and maresin 1 to leukotriene B4 ratios in the plasma.(2019) Perucci, Luiza Oliveira; Santos, Talita Adriana Pereira dos; Santos, Patrícia Campi; Teixeira, Lívia Cristina Ribeiro; Alpoim, Patrícia Nessralla; Gomes, Karina Braga; Sousa, Lirlândia Pires; Dusse, Luci Maria Sant'Ana; Silva, André Talvani Pedrosa daProblem: Omega-3 and omega-6 fatty acids can be endogenously converted into mediators with pro-inflammatory (eg, leukotriene B4/LTB4) or anti-inflammatory/ pro-resolving activities (eg, resolvin D1/RvD1 and maresin 1/MaR1). Recent data indicate an imbalance of LTB4 and MaR1 levels in pre-eclampsia (PE), but the relative production of these mediators, including RvD1, and the role of these mediators in the disease pathogenesis remain unclear. Therefore, this study aimed to investigate the plasma levels of LTB4, RvD1, and MaR1 in pregnant women with or without PE and non-pregnant controls and their association with clinical/laboratory parameters of PE women. Method of study: LTB4, RvD1, and MaR1 plasma levels were measured by competitive enzyme immunoassay in 19 non-pregnant, 20 normotensive pregnant, and 21 PE women. Results: Plasma concentrations of LTB4 were higher and RvD1 were lower in PE women than in normotensive pregnant women, who presented higher levels of LTB4 and similar levels of RvD1 to non-pregnant women. MaR1 levels did not differ among the groups. Pre-eclampsia women had decreased RvD1/LTB4 and MaR1/LTB4 ratios. Considering only the PE group, positive correlations were observed among all the mediators tested, between LTB4 and white blood cell count and between RvD1 and creatinine levels. However, all lipid mediators correlated negatively with body mass index before pregnancy. LTB4 also correlated negatively with maternal age. Conclusion: Our findings suggest that the PE state results in systemic overproduction of LTB4 in relation to RvD1 and MaR1, and that these lipid mediators may be involved with the disease pathogenesis.
