Item Alterations in the proteomic composition of Serratia marcescens in response to manganese (II).(2018) Queiroz, Pollyana Santos; Ruas, France Anne Dias; Barboza, Natália Rocha; Borges, William de Castro; Cota, Renata Guerra de SáBackground: Proteomics is an important tool for the investigation of dynamic physiological responses of microbes under heavy metal stress. To gain insight into how bacteria respond to manganese (II) and identify the proteins involved in Mn (II) oxidation, the shotgun proteomics approach was applied to a potential Mn (II)-oxidizing Serratia marcescens strain cultivated in the absence and presence of Mn (II). Results: The LG1 strain, which grew equally well in the two conditions, was found to express a set of proteins related to cellular processes vital for survival, as well as proteins involved in adaptation and tolerance to Mn (II). The multicopper oxidase CueO was identified, indicating its probable participation in the Mn (II) bio-oxidation; however, its expression was not modulated by the presence of Mn (II). A set of proteins related to cell and metabolic processes vital to the cells were downregulated in the presence of Mn (II), while cell membrane-related proteins involved in the maintenance of cell integrity and survival under stress were upregulated under this condition. Conclusions: These findings indicate that the LG1 strain may be applied successfully in the bioremediation of Mn (II), and the shotgun approach provides an efficient means for obtaining the total proteome of this species.Item High manganese tolerance and biooxidation ability of Serratia marcescens isolated from manganese mine water in Minas Gerais, Brazil.(2017) Barboza, Natália Rocha; Morais, Mônica Mendes Cordeiro Araújo; Queiroz, Pollyana Santos; Amorim, Soraya Sander; Cota, Renata Guerra de Sá; Leão, Versiane Albisanganese is an important metal for the maintenance of several biological functions, but it can be toxic in high concentrations. One of the main forms of human exposure to metals, such as manganese (Mn), is the consumption of solar salt contaminated. Mn-tolerant bacteria could be used to decrease the concentration of this metal from contaminated sites through safer environmental-friendly alternative technology in the future. Therefore, this study was undertaken to isolate and identify Mn resistant bacteria from water samples collected from a Mn mine in the Iron Quadrangle region (Minas Gerais, Brazil). Two bacterial isolates were identified as Serratia marcescens based on morphological, biochemical, 16S rDNA gene sequencing and phylogeny analysis. Maximum resistance of the selected isolates against increasing concentrations of Mn(II), up to 1200 mg L-1 was determined in solid media. A batch assay was developed to analyze and quantify the Mn removal capacities of the isolates. Biological Mn removal capacities of over 55% were detected for both isolates. Whereas that mechanism like biosorption, precipitation and oxidation could be explaining the Mn removal, we seek to give an insight into some of the molecular mechanisms adopted by S. marcescens isolates. For this purpose, the following approaches were adopted: leucoberbelin blue I assay, Mn(II) oxidation by cell-free filtrate and electron microscopy and energy-dispersive X-ray spectroscopy analyses. Overall, these results indicate that S. marcescens promotes Mn removal in an indirect mechanism by the formation of Mn oxides precipitates around the cells, which should be further explored for potential biotechnological applications for water recycling both in hydrometallurgical and mineral processing operations.Item A new application for Klebsiella oxytoca in bioremediation : treatment of manganese-laden wastewaters.(2018) Barboza, Natália Rocha; Morais, Mônica Mendes Cordeiro Araújo; Santos, Pricila Almeida; Queiroz, Pollyana Santos; Leão, Versiane Albis; Cota, Renata Guerra de SáBioremediation is a method of removing manganese from wastewaters, and many bacterial species with a role in manganese bioremediation have been identified over several decades. Using K medium with Mn(II), a bacterial consortium was obtained from water sample collected from a mine drainage in southeastern Brazil (Minas Gerais, Brazil), and the isolates were identified as Klebsiella oxytoca using both biochemical and 16S rRNA phylogenetic analysis. Among such isolates, sample SA8 was tested for its manganese removal capability during a week-long experiment in K medium amended with manganese sulfate. Growth of isolate SA8 produced an increase in pH concomitantly to Mn(II) removal, which was not observed in the abiotic control. Manganese removal occurred through oxidation as confirmed by reaction with leucoberbelin blue, and therefore, a new potential application for K. oxytoca in mine water bioremediation is proposed.Item Rich growth medium promotes an increased on Mn(II) removal and manganese oxide production by Serratia marcescens strains isolates from wastewater.(2018) Queiroz, Pollyana Santos; Barboza, Natália Rocha; Morais, Mônica Mendes Cordeiro Araújo; Leão, Versiane Albis; Cota, Renata Guerra de SáThis study investigated the performance of two different strains of Serratia marcescens on the manganese (II) removal and the effects of the nutrient-rich (NB medium) and nutrient-poor culture (K medium) media on this process. In the NB medium, the pigmented LG1 strain and the non-pigmented CL11 strain exhibited improved growth and higher Mn(II) tolerance (0–2000 mg L−1 ). In addition, improved Mn(II) removal (64.25%) and increased formation of Mn oxides were observed in this medium, especially for the LG1 isolate. EELS analysis revealed Mn inside the cell for LG1 and EDX analysis revealed Mn outside of the cell for CL11, indicating that the two strains remove Mn(II) via distinct mechanisms. The biooxidation of Mn by the CL11 strain may involve indirect mechanisms which alter the pH of the medium, whereas the LG1 strain may use a direct mechanism for the biooxidation mediated by cellular components such as intracellular proteins. These results demonstrate the biotechnological potential of the two strains. In addition, is the first time the high potential of the NB medium for Mn removal is demonstrated. This medium was found to greatly influence the Mn removal and oxide formation, especially in the case of the pigmented LG1 isolate.Item Serratia marcescens resistente ao manganês e estratégias para biorremediação de ambientes contaminados.(2018) Queiroz, Pollyana Santos; Cota, Renata Guerra de Sá; Góes Neto, Aristóteles; Cota, Renata Guerra de Sá; Babá, Élio Hideo; Andrade, Milton Hércules Guerra deO manganês (II) solúvel que tem sido associado a uma crescente contaminação de corpos aquáticos e problemas de saúde humana devido à sua toxicidade. Na busca por estratégias ecologicamente adequadas para a sua remoção, podemos destacar os isolados de Serratia marcescens, que teve seu o potencial demonstrado na remoção de altas concentrações desse metal. Como forma de melhorar a eficiência de remoção de Mn (II) dessa espécie, um meio de cultura rico em nutrientes (meio NB) suplementado com Mn (II) foi comparado com um meio pobre em nutrientes (meio K) para avaliar a sua influência no processo de remoção por duas diferentes cepas, uma não pigmentada (CL11) e outra pigmentada (LG1) que ainda não tinha sido estudada. Uma vez que um meio rico pode favorecer um maior crescimento bacteriano, uma maior remoção de Mn (II) pode ocorrer. No meio NB, a cepa LG1 e a cepa CL11 exibiram um melhor crescimento e maior tolerância ao Mn (II) (0-2000 mg L − 1). Além disso, uma melhor remoção de Mn (II) (64,25%) e o aumento da formação de óxidos de Mn foram observados neste meio, especialmente para o isolado LG1. A análise de EELS revelou Mn no interior da célula de LG1 e a análise de EDX revelou Mn fora da célula de CL11, indicando que as duas cepas removem Mn (II) através de mecanismos distintos. A bioxidação de Mn pelo isolado CL11 parece envolver mecanismos indiretos que alteram o pH do meio, enquanto o isolado LG1 parece usar um mecanismo direto para a biooxidação mediada por componentes celulares, como proteínas intracelulares. É a primeira vez que o alto potencial do meio NB para remoção de Mn é demonstrado. Este meio influenciou em uma melhor remoção de Mn e na formação de óxidos, especialmente no caso do isolado LG1 pigmentado. Como a cepa LG1 apresentou um potencial diferenciado na bio-oxidação de Mn (II), o seu foi proteoma foi analisado na ausência e presença de Mn (II) através da abordagem shotgun, para obter informações sobre como as bactérias respondem a esse metal e identificar as proteínas envolvidas na sua oxidação. A cepa LG1, que cresceu igualmente bem nas duas condições, expressou um conjunto de proteínas relacionadas aos processos celulares vitais para a sobrevivência, bem como proteínas envolvidas na adaptação e tolerância ao Mn (II). A multicobre oxidase CueO foi identificada, indicando sua provável participação na bioxidação de Mn (II), no entanto, sua expressão não foi modulada pela presença desse metal. Um conjunto de proteínas relacionadas aos processos celulares e metabólicos vitais para as células foram reguladas negativamente na presença de Mn (II), enquanto as proteínas relacionadas à membrana celular envolvidas na manutenção da integridade celular e sobrevivência sob estresse foram upreguladas sob essa condição. Este estudo permitiu obter o primeiro proteoma total dessa espécie nas condições de ausência de presença de Mn (II). O pigmento prodigiosina sintetizado pela cepa LG1 foi caracterizado e foi possível observar o efeito de elevadas concentrações de Mn (II) na sua produção e o seu efeito protetor para a cepa nessas condições. Entretanto é necessário estudar a relação direta da prodigiosina com a remoção de Mn (II). Em conjuntos, estes resultados demonstram o potencial biotecnológico de isolados de S. marcescens na biorremediação de Mn (II) e sugerimos a sua utilização juntamente com o meio NB em grandes biorreatores contínuos para o tratamento de efluentes contaminados com Mn. Além disso, a lista de proteínas expressas identificadas pela análise proteômica pode ser usada como uma ferramenta em experimentos futuros para validar esses achados.
