Heavy metals biomonitoring on the environment was this study objective using A. viridis compared with Perna perna (mollusk bivalve). The area studied included Ilha Pituã in Vila Velha – ES and Ilha do Boi and Camburi Garden Beach in Vitoria - ES. METHODS: The sample collection comprised seawater, mussel (P. perna) and sponge (A. viridis). Al, As, Ba, Cr, Cu, Fe, Pb, Mn, K, Ti and Zn were analyzed using ICP OES. RESULTS For metal studied Al, Ba, Cr, Fe and Mn presented greater accumulation in A. viridis than in P. perna. The metal concentrations detected in seawater, sponge, and mussel could be related using PCA with 79.26% total variance demonstrating that A. viridis bioaccumulated more metals than P. perna. CONCLUSION: Sponge metal accumulation reflected not only spatial metal variation but also seasonal changes.Item Efeitos antiproliferativos de naftoquinonas em linhagens tumorais de bexiga.(2022) Ferreira, Gabriel Monteze; Brandão, Geraldo Célio; Silva, Glenda Nicioli da; Brandão, Geraldo Célio; Endringer, Denise Coutinho; Dolabela, Maria Fâni; Souza, Gustavo Henrique Bianco de; Cardoso, Leonardo MáximoAs quinonas são compostos químicos produzidos a partir da oxidação de fenóis, Uma de suas principais características é a presença de dois grupos carbonílicos que formam um sistema conjugado com pelo menos duas ligações duplas em um anel de seis carbonos. Entre as quinonas, as naftoquinonas se destacam como potenciais agentes antitumorais. Com base neste histórico, oito naftoquinonas da biblioteca de moléculas do Laboratório de Química Medicinal e Bioensaios da Escola de Farmácia da UFOP tiveram suas citotoxicidades testadas pelo método colorimétrico do MTT (brometo de [3-(4,5-dimetiltiazol2yl)-2,5-difenil tetrazolium]) em linhagens de carcinoma urotelial. A nafto [2,3-b] tiofen-4,9-quinona (NQ1), 8-metoxi-α-lapachona (NQ3) e a lausona (NQ8) apresentaram melhores índices de seletividade para células de carcinoma urotelial e, portanto, foram selecionadas para outros ensaios. A NQ1 apresentou índice de seletividade de 7,32; 18,16 e 10,55 respectivamente para as linhagens RT4, 5637 e T24, enquanto a NQ3 e a NQ8 apresentaram índice de seletividade respectivamente de 19,5 e 28,0 para a linhagem T24. Os cálculos foram feitos utilizando os valores de citotoxicidade obtidos para a linhagem não tumoral MRC-5 (fibroblasto humano). No ensaio de sobrevivência clonogênica, a maior concentração testada de NQ1 (2,5 µg/mL) reduziu 76, 52 e 48% a formação de clones para as linhagens RT4, 5637 e T24 respectivamente. A NQ3 reduziu 70,5% e a NQ8 reduziu 87,1% a formação de clones para a linhagem T24. Observou-se também que a NQ1 alterou a morfologia das três linhagens testadas e a NQ3 alterou a morfologia da T24, onde foi possível observar a presença de células alongadas. As três moléculas também apresentaram o efeito de diminuição no processo de migração celular. As três naftoquinonas alteraram o ciclo celular e induziram significativamente a produção de espécies reativas de oxigênio (EROs), que estão ligadas a danos no DNA. Em conclusão, as naftoquinas estudadas apresentaram potencial efeito antiproliferativo em linhagem tumoral de câncer de bexiga, interferindo no ciclo e na migração celular celular, possivelmente por meio de estresse oxidativo.Item Endothelium-dependent vasodilation induced by Hancornia speciosa in rat superior mesenteric artery.(2007) Ferreira, Herick Campos; Serra, Carla Penido; Endringer, Denise Coutinho; Lemos, Virgínia Soares; Braga, Fernão Castro; Côrtes, Steyner de FrançaThe vasodilator effect of the ethanolic extract of leaves from Hancornia speciosa Gomes (HSE) was evaluated in superior mesenteric artery rings. HSE produced a concentration-dependent vasodilation (IC50 ¼ 10.874.0 mg/mL) in arterial rings pre-contracted with phenylephrine, which was completely abolished in endothelium-denuded vessels. Endothelium-dependent vasodilation induced by HSE was strongly reduced by L-NAME (100 mM), a nitric oxide (NO) synthase inhibitor, but neither by atropine, a muscarinic receptor antagonist (1 mM), nor by indomethacin (10 mM), a cyclooxygenase inhibitor. In rings pre-contracted with 80mM KCl, the vasodilator effect of HSE was shifted to the right and was completely abolished in the presence of L-NAME (100 mM). Similar effects were obtained in mesenteric rings pre-contracted with phenylephrine in the presence of KCl 25mM alone or in addition to 100 mM L-NAME.In addition, BaCl2 (1mM) dramatically reduced the vasodilation induced by HSE. Together, these findings led usv to conclude that HSE induces an endothelium-dependent vasodilation in rat mesenteric artery, by a mechanism dependent on NO, on the activation of potassium channels and endothelium-derived hyperpolarizing factor release. Rutin, identified as a major peak in the HPLC fingerprint obtained for HSE, might contribute for the observed vasodilator effect, since it was able to induce an endothelium-dependent vasodilation in rat superior mesenteric arteries.Item Mangifera indica L. as airborne metal biomonitor for regions of the state of Espírito Santo (Brazil).(2020) Castro, Leonardo Zago de; Silva, Thaís R. Bioindicators present an alternative method of monitoring air quality. This work proposes to evaluate the Mangifera indica L. leaves as a biomonitor for air quality. Quantification of aluminum (Al), barium (Ba), calcium (Ca), chromium (Cr), copper (Cu), iron (Fe), potassium (K), manganese (Mn), magnesium (Mg), sodium (Na), nickel (Ni), sulfur (S), strontium (Sr), titanium (Ti), and zinc (Zn) were determined in leaves of M. indica using optical emission spectrometer inductively coupled plasma (ICP-OES). The correlogram analyses demonstrated a strong positive correlation between Al and Fe. Contamination of the soil by vehicles and agricultural chemicals, in synergy with the influence of the winds, may be considered as a source of contamination. Enrichment factor (EF) index was used to distinguish between natural and anthropogenic sources. Detection of Mn and Cu could be associated with anthropogenic influence, demonstrating M. indica as a feasible tool to biomonitor air quality.
